Called Duke Nukem Begins it was set to be a Duke origin story developed by Gearbox Software alongside 2K and 3D Realms. This 2008 animated vertical slice was created by a company called Janimation and features plenty of third-person action, alongside looks at potential co-op. None of it is actual game footage, but a very cool look at a Duke reboot that was ultimately cancelled.
This video comes from Gregor Punchatz who writes, "I headed up this project, as the director at Janimation, and did a lot of the character development, including the design and modeling of Duke. We poured everything we had into making the most awesome cinematic Janimation had ever created. But unfortunately, we got word the game was canceled due to the legal issues with Duke Nukem at the time."

It's a pretty slick trailer depicting what the game might have looked like had it been made -- including, as mentioned above, four Dukes running around in different coloured tees shooting alien bad guys.
